I am playing against Tottenham at White Hart Lane right now, and i'm in big trouble. I was very lucky, when i only lost 3-2. They had 23 shot and i had 8. Does the tactic not work against the big teams? Because the tactic is effective against the sub-teams and smaller teams.

Since FM is a simulation game, sometimes bad result could happen.

I just tried to have 4 matches using Barca, and putting messi as left striker was very good.
And using Vermaelen instead of Mascherano for CB was good since Mascherano's jumping and heading are too bad.

I plugged your "wrong" version and got a 7-1 away win followed by a 9-0 home win immediately... Seems to me that version is still decent

(however I did remove the "close down much more" PI on the central defender, to try and mend for me being overrun through the middle of my defence in this tactic)
